TITLE:Coordinating Photorespiration and Flowering by a LncRNA-mediated Nuclear-cytoplasmic Partition of a Peroxisomal Enzyme
ABSTRACT:
The photorespiration is a metabolic repair system that enables the CO2 fixation enzyme Rubisco to operate sustainably in the presence of oxygen. Despite the extensive research on the biochemical processes of photorespiration, the cellular regulatory mechanisms of core enzymes involved in the photorespiration remain unexplored. Here we found that an Arabidopsis lncRNA, functions as a scaffold to mediate the interaction between the nuclear-cytoplasmic shuttle protein and hydroxypyruvate reductase (HPR1), a core enzyme in the photorespiration pathway. Notably, we observed that light/dark transitions induce the nuclear-cytoplasmic translocation of HPR1. Furthermore, lncRNA and the nuclear-cytoplasmic shuttle protein modulate the exporting of HPR1 out of nucleus during the transition from darkness to light, which is essential for the normal photorespiration process. Moreover, the nuclear HPR1and BLIL2 module regulates flowering by affecting the splicing of FLC sense transcript. Our findings reveal that lncRNA and the nuclear-cytoplasmic shuttle protein regulate the nuclear-cytoplasmic partition of HPR1, thus coordinate the photorespiration and flowering pathways.
